Summary

Hydrogen Peroxide (H2O2) is a highly reactive species normally produced in cells by both enzymatic and non-enzymatic pathways. The reaction of superoxide disputase (SOD) with superoxide radicals within cells and in surrounding areas results in the uncontrolled formation of H2 O2. Granulocytes also produce H2O2 via metabolic processes as an immunological response to foreign invaders. While the metabolic production of H2O2 by granulocytes is controlled at the immunological level, the production of H2O2 as a by-product of oxidative stres is not wellregulated and thus cells must rely on the enzymes catalase and glutathione peroxidase to convert H2O2 into harmless water and oxygen. As a by-product of the SOD oxidative stress response and as an integral part of normal inflammatory response to infection, understanding the impact of H2O2 on various model systems hes become an important facet of studying many pathophysiological processes.

The NWLSS™ Hydrogen Peroxide assay is based on the oxidation of ferrous ions to ferric ions by peroxides wherein the ferric ions react with the indicator dye xylenol orange to produce a purple colored complex measureable at 560-590 nm. Sorbitol is included to amplify the color intensity. Reaction of samples with and without catalase enzyme provides the means to specifically quantify H2O2 by difference in sample unknowns.
